ID 1.1.1.375 DE L-2-hydroxycarboxylate dehydrogenase [NAD(P)(+)]. AN lactate/malate dehydrogenase. CA (1) a (2S)-2-hydroxycarboxylate + NAD(+) = a 2-oxocarboxylate + H(+) + CA NADH. CA (2) a (2S)-2-hydroxycarboxylate + NADP(+) = a 2-oxocarboxylate + H(+) + CA NADPH. CC -!- The enzyme from the archaeon Methanocaldococcus jannaschii catalyzes CC the reversible oxidation of (2R)-3-sulfolactate and (S)-malate to CC 3-sulfopyruvate and oxaloacetate, respectively (note that (2R)-3- CC sulfolactate has the same stereochemical configuration as (2S)-2- CC hydroxycarboxylates). CC -!- The enzyme can use both NADH and NADPH, although activity is higher CC with NADPH. CC -!- The oxidation of (2R)-3-sulfolactate was observed only in the CC presence of NADP(+). CC -!- The same organism also possesses an NAD(+)-specific enzyme with CC similar activity, cf. EC 1.1.1.337. DR Q60176, MDH_METJA ; //
