CA [thioredoxin]-disulfide + a 2'-deoxyribonucleoside 5'-diphosphate + H2O = CA [thioredoxin]-dithiol + a ribonucleoside 5'-diphosphate. CC -!- This enzyme is responsible for the de novo conversion of CC ribonucleoside diphosphates into deoxyribonucleoside diphosphates, CC which are essential for DNA synthesis and repair. CC -!- There are three types of this enzyme differing in their cofactors. CC -!- Class Ia enzymes contain a diiron(III)-tyrosyl radical, class Ib CC enzymes contain a dimanganese-tyrosyl radical, and class II enzymes CC contain adenosylcobalamin. CC -!- In all cases the cofactors are involved in generation of a transient CC thiyl (sulfanyl) radical on a cysteine residue, which attacks the CC substrate, forming a ribonucleotide 3'-radical, followed by water CC loss to form a ketyl (alpha-oxoalkyl) radical. CC -!- The ketyl radical is reduced to 3'-keto-deoxynucleotide concomitant CC with formation of a disulfide anion radical between two cysteine CC residues. CC -!- A proton-coupled electron-transfer from the disulfide radical to the CC substrate generates a 3'-deoxynucleotide radical, and the the final CC product is formed when the hydrogen atom that was initially removed CC from the 3'-position of the nucleotide by the thiyl radical is CC returned to the same position.
