ID 1.14.99.66 DE [histone-H3]-N(6),N(6)-dimethyl-L-lysine(4) FAD-dependent demethylase. AN lysine-specific histone demethylase 1. CA 2 A + 2 H2O + N(6),N(6)-dimethyl-L-lysyl(4)-[histone H3] = 2 AH2 + CA 2 formaldehyde + L-lysyl(4)-[histone H3]. CC -!- The enzyme specifically removes methyl groups from mono- and CC dimethylated lysine(4) of histone 3. CC -!- During the reaction the substrate is oxidized by the FAD cofactor of CC the enzyme to generate the corresponding imine, which is subsequently CC hydrolyzed in the form of formaldehyde. CC -!- The enzyme is similar to flavin amine oxidases, and differs from all CC other known histone lysine demethylases, which are iron(II)- and CC 2-oxoglutarate-dependent dioxygenases. CC -!- The physiological electron acceptor is not known with certainty. In CC vitro the enzyme can use oxygen, which is reduced to hydrogen CC peroxide, but generation of hydrogen peroxide in the chromatin CC environment is unlikely as it will result in oxidative damage of DNA.
