ID 3.2.1.220 DE ipecoside beta-D-glucosidase. AN 6-O-methyl-deacetylisoipecoside beta-glucosidase. AN IpeGlu(1). CA (1) H2O + ipecoside = D-glucose + ipecoside aglycone. CA (2) H2O + N-deacetylipecoside = D-glucose + N-deacetylipecoside aglycone. CA (3) 6-O-methyl-N-deacetylipecoside + H2O = 6-O-methyl-N-deacetylipecoside CA aglycone + D-glucose. CC -!- The enzyme, isolated from the roots of the plant Carapichea CC ipecacuanha, preferentially hydrolyzes glucosidic ipecoside alkaloids CC except for their lactams, but shows poor or no activity toward other CC substrates. CC -!- IpeGlu(1) activity is extremely poor toward 7-O-methyl and 6,7- CC O,O-dimethyl derivatives. However, 6-O-methyl derivatives are CC hydrolyzed as efficiently as non-methylated substrates. CC -!- IpeGlu(1) accepts both 1alpha(S)-N-deacetylisoipecoside and 1beta(R)- CC N-deacetylipecoside epimers as substrate, with preference for the CC 1beta(R)-epimer. CC -!- 6-O-methyl-N-deacetylisoipecoside is an intermediate in the CC biosynthesis of the medicinal alkaloid emetine. //
